Description
Creative Biolabs provides large-scale non-GMP plasmid DNA manufacturing services for in vivo, in vitro and pre-clinical applications. While most of the plasmid DNA we produce is used for gene therapy research and DNA vaccines, we also manufacture DNA for other applications, including transient tranfection of mammalian cells, toxicology studies and as plasmid-based diagnostic reagents.
A proprietary procedure covering bacterial cell fermentation and plasmid DNA purification was well established to produce ultrapure plasmid DNA at extremely low endotoxin level [<2 EU/mg] and great homogeneity [≥ 99 % super-coiled with linear and nicked forms removed].
Of note, we are able to produce unstable plasmid DNA, which is the most challenging issue in the field. We have created a bacterial strain that was gene-engineered to remove genes involved in the rearrangement and deletion of DNA repeats. This bacterial strain has both the UV repair system and the SOS repair pathway altered, which are involved in repairing DNA lesions. In addition, a certain set of DNA recombination proteins, including recA gene, are mutated. This special bacterial strain significantly reduces homologous DNA recombination/rearrangement and DNA deletion, thus guaranteeing the integrity of unstable plasmid DNA of large sizes in manufacturing.
